Caractérisation moléculaire du gène RHD chez les donneurs de sang présentant le phénotype Rh DEL au Centre National de Transfusion Sanguine de Bamako au Mali. (French)

Introduction: DEL phenotype is a rare Rh variant characterized by a very low level of D antigen expression at the surface of red blood cells. The frequency of RH DEL is unknown in Mali. The purpose of this study was to characterize the RHD gene using conventional PCR and to verify the presence of the RHD 1227A allele in blood donors with the DEL phenotype at the National blood transfusion Center in Bamako. Methods: after detection of the Rh DEL by adsorption-elution technique, DNA was isolated and amplified by conventional PCR using primers specific for the RHD gene and the RHD 1227A allele. SPSS Statistics Version 26 was used to analyze data. Results: a total of 365 RhD-negative blood donors from the CNTS in Bamako were recruited for this study. There was a prevalence in men (90,4%); the age group 26-39 years was the most represented, with a frequency of 52,9%. The frequency of Rh DEL phenotype was 7.1% and blood group O was the most represented (38.6%) in the population as a whole. CCEE phenotype accounted for 61,5% of the DEL phenotypes. PCR-SSP showed amplification of the 10 exons of the RHD gene, with the presence of (C) haplotype. RHD 1227A allele was present in all RH DEL samples. Conclusion: RH-DEL phenotype carrying the RHD1227A allele with an intact RHD gene was found in blood donors in Mali. [ABSTRACT FROM AUTHOR]

Introduction: le Rh DEL est un variant rare du système RH qui s'exprime très faiblement à la surface des hématies. La fréquence du Rh DEL est méconnue au Mali. L'objectif de cette étude était de caractériser le gène RHD par PCR classique et vérifier la présence de l'allèle RHD 1227A chez les donneurs de sang présentant le phénotype DEL au Centre National de Transfusion sanguine de Bamako. Méthodes: après une confirmation du Rh DEL par adsorption/élution, l'ADN des participants a été isolé et amplifié par PCR classique en utilisant des amorces spécifiques au gène RHD et l'allèle RHD 1227A. Le logiciel SPSS version 26 a été utilisé pour l'analyse des données. Résultats: un total de 365 donneurs de sang RhD négatif du CNTS de Bamako, ont été recrutés dans cette étude. Le sexe masculin était plus fréquent avec 90,4%; la tranche d'âge 26-39 ans était la plus représentée avec une fréquence de 52,9%. La fréquence du phénotype Rh DEL était de 7,1% et le groupe sanguin O était le plus représenté avec 38,6% dans l'ensemble de la population. Le phénotype Ccee représentait 61,5% des phénotypes DEL. La PCR-SSP a montré une amplification des 10 exons du gène RHD avec la présence de l'haplotype (C). L'allèle RHD 1227A était présent chez tous les échantillons Rh DEL.
