Rapid screening and analysis of alcohol dehydrogenase binders from Glycyrrhiza uralensis root extract using functionalized magnetic nanoparticles coupled with HPLC-MS/MS.

Alcohol dehydrogenase (ADH) is a key enzyme that converts ethanol into acetaldehyde. Its binders could be used to efficiently treat human toxic alcohol poisoning and suppress the acetaldehyde accumulation in alcohol hypersensitive alcoholics. In the present study, a new assay based on ADH-functionalized magnetic nanoparticles coupled with high-performance liquid chromatography - tandem mass spectrometry (HPLC-MS/MS) was developed for the rapid screening and identification of ADH binders. ADH was immobilized on silica-coated Fe3O4 magnetic nanoparticles via covalent bonds and the synthesized nanoparticles were characterized by TEM, XRD, FT-IR, and VSM. The amount of bound ADH onto magnetic nanoparticles was 80.6 μ/mg. The optimum temperature and pH for the enzymatic activity of immobilized ADH were 25℃ and 7.0, respectively. The relative activity of immobilized ADH remained 68.14% after 10 times of recycle, which exhibited good durability. Nine compounds with ADH-binding activity were screened and identified in Glycyrrhiza uralensis root extracts, among which seven compounds were first screened and identified. L'alcool déshydrogénase (ADH) est une enzyme essentielle qui convertit l'éthanol en aldéhyde. Les inhibiteurs de l'ADH pourraient être utilisés pour traiter efficacement l'intoxication alcoolique chez l'homme et supprimer l'accumulation d'acétaldéhyde chez les alcooliques hypersensibles a l'alcool. La présente étude décrit l'élaboration d'un nouvel essai fondé sur des nanoparticules magnétiques fonctionnalisées avec de l'ADH conjuguées a la chromatographie en phase liquide a haute performance couplée à la spectrométrie de masse en tandem (HPLC-MS/MS) pour la détection et l'identification rapides des inhibiteurs de l'ADH. On a immobilisé l'ADH sur des nanoparticules magnétiques de Fe3O4 enrobées de silice par des liaisons covalentes et caractérisé les nanoparticules synthétisées par MET, DRX, IRTF et VSM. La quantité d'ADH liée aux nanoparticules magnétiques était de 80,6 μ/mg. Les valeurs de température et de pH optimales pour l'activité enzymatique de l'ADH immobilisée étaient 25℃ et 7,0, respectivement. L'activité relative de l'ADH immobilisée était encore de 68,14% après 10 recyclages, ce qui témoigne d'une bonne durabilité. Neuf composés présentant une activité de liaison avec l'ADH ont été décelés et identifiés dans des extraits de racine de Glycyrrhiza uralensis, parmi lesquels sept l'ont été pour la première fois. Les résultats expérimentaux prouvent que la méthode proposée permettrait de déceler et d'identifier rapidement les inhibiteurs de l'ADH dans des mélanges complexes. [ABSTRACT FROM AUTHOR]
